The right size in Ralph Lauren depends on two main things: the size on the label, and the fit of the piece.

The roomier fit. It's cut with more room through the chest and waist, for a classic, relaxed look.
If you like a shirt to sit a little loose rather than close to the body, this is usually the better fit.
Custom Slim Fit
Ralph Lauren's in-between fit.
It's more tailored than Classic Fit, mainly through the waist, but still comfortable rather than tight.
Slim Fit
The narrowest, most tailored fit.
It sits closer to the body, with less volume through the chest and waist. For anyone who prefers a slimmer, sharper look.
How to choose your size
If you already have a Ralph Lauren shirt that fits you well, check the size and fit on its label. That's the best reference point for the pieces on the site.
For example, if you usually wear M in Custom Slim Fit, looking for that same combination of size and fit will give you a better starting point than the size alone.
If you move between fits, bear in mind that the same size can feel different on the body.
Fit by garment type
Polo shirts
In a polo, the fit makes a big difference. Classic Fit will feel roomier, while Slim Fit sits closer to the body.
Button-downs
With button-downs, pay particular attention to the fit. A tailored model can feel narrower through the chest and waist even when the size matches another of the brand's shirts.
Knitwear
